Investigating the molecular mechanisms underlying the anti‐CRISPR function of AcrIIA13b protein

Anti‐CRISPR protein AcrIIA13b inhibits CRISPR–Cas9 function by preventing target DNA engagement. The Cas9–sgRNA ribonucleoprotein (RNP) normally recognizes the protospacer adjacent motif (PAM) duplex to initiate DNA cleavage. AcrIIA13b binding to the PAM‐interacting WED–PI domain blocks this recognition, leaving the target DNA intact.

Oxygen‐independent expression of HIF‐1α during the cell cycle in hepatocellular carcinoma cells controls essential metabolic pathways under normoxia

In Huh7 cells, HIF‐1α is essential as it maintains the expression of proteins involved in glycolysis and steroid/cholesterol biosynthesis both under normoxia and hypoxia. On the other hand, in HeLa cells, these pathways are induced by HIF‐1α only under hypoxia.

Cell‐surface CLDN‐5 protein levels can be evaluated using a probe that can bind to the extracellular domains of CLDN‐5. A probe derived from Clostridium perfringens enterotoxin allows us to isolate cells with high CLDN‐5 protein levels from a knockout cell library.
