Cutting Sections of Paraffin-Embedded Tissues
Cold Spring Harbor Protocols, 2008INTRODUCTIONThis protocol describes the sectioning of tissues embedded in paraffin blocks. Paraffin sections require extensive fixation and processing steps but provide superior morphology compared with other sectioning methods. Sectioning paraffin blocks requires experience and should be learned from an experienced researcher, if possible.

Softening Paraffin-Embedded Plant Tissues
Stain Technology, 1953Soaking paraffin-embedded plant specimens 2–3 days at 37°C. in a mixture of glycerol, 10 ml., Dreft, 1 g., and water 90 ml. is an effective means of softening them prior to sectioning. One side of the paraffin block must be pared away to expose the tissue before immersion in the softening solution.

HOPE fixation: a novel fixing method and paraffin-embedding technique for human soft tissues.
Pathology, Research and Practice, 2001We have developed a novel method for tissue fixation, including subsequent paraffin-embedding and sectioning, that allows the complete pathological analysis of all types of human soft tissues.

Improvement of the Butyl Methacrylate-Paraffin Embedment
Stain Technology, 1983The excellent butyl methacrylate-paraffin method as an embedment for light microscopy has been technically improved. More uniform and reproducible polymerization has been obtained by using a vacuum oven to degas the polymerizing mixture and to replace the air with nitrogen at 650 Torr. The amount of benzoyl peroxide required must be determined for each

Embedding Paraffins for Eye Microtomy
Journal of Histotechnology, 1977Abstract Embedding paraffins commonly used for eye microtomy are discussed. A relatively new paraffin product may be used also for eye microtomy and light microscopy preparations of excellent quality.

A Sample-Grouping Technique for Paraffin Embedments
Stain Technology, 1983A technique is described which facilitates histological preparation of multiple tissue specimens for light microscopy. The procedure enables the investigator to separate and label identifiable subgroups from a larger number of specimens in one histological section. After standard fixation, murine esophagi were arranged longitudinally and secured within

N-Butyl methacrylate and paraffin as an embedding medium for light microscopy.
Stain Technology, 1978A method of tissue embedding using n-butyl methacrylate and paraffin is described. Following alcohol dehydration and infiltration with the methacrylate monomer, tissues are embedded in gelatin capsules in a mixture consisting of 3.5 g of paraffin for ...

Orienting and Embedding Small Objects in Paraffin
Stain Technology, 1956A beaker filled with ice cubes and water is covered with a plate of sheet metal. Over this is placed a paper embedding boat full of molten paraffin, and an inch or two above it is held a 60-watt soldering iron. The height of the soldering iron is then adjusted so that the paraffin at the lower half of the embedding dish becomes semisolid and the upper ...
