Mechanical Activation of Piezo1 Drives Osteoarthritis Through Kdm5c‐Mediated Epigenetic Silencing

Excessive mechanical stress activates Piezo1, triggering Ca2+‐dependent cytoskeletal forces that deform the nucleus and reduce H3K4me3. Kdm5c demethylates H3K4me3 at Col2a1 and Runx3 promoters. Cancer vaccines face limitations in pancreatic cancer due to insufficient tumor antigens. This ionic liquid ILvax via local tumor delivery triggers robust cDC1‐dependent systemic anti‐tumor immunity. It boosts endogenous CD8+ T cells and optimizes adoptive TCR‐T function, providing a promising synergistic therapeutic strategy.

Cholesterol‐enriched plasma membranes in hepatocellular carcinoma impede drug penetration. Cholesterol (+)‐liposomes act as membrane‐specific detergents, extracting cholesterol and reducing barrier function without cytotoxicity. Following endocytosis, cholesterol transfers from endosomes to liposomes and is metabolized in the ER.

Targeting a Myeloid–Regulatory B Cell Network Reverses Immune Paralysis in Periprosthetic Joint Infections

Periprosthetic joint infection establishes a sophisticated immunosuppressive network between CXCR4+ PMN‐MDSCs and Bregs, inducing profound CD8+ T cell paralysis. Alendronate effectively disrupts this CXCR4+ PMN‐MDSC–Breg axis by targeting STAT3, thereby restoring local immune surveillance.
