ATP Hydrolysis by α‐Synuclein Amyloids is Mediated by Enclosing β‐Strand

open access: yesAdvanced Science, EarlyView.
Pathological amyloids have been considered chemically inert until recently. Here it is shown that α‐synuclein amyloids catalyze the hydrolysis of the universal energy molecule, ATP. The cryo‐EM structure of the complex reveals an additional β‐strand (purple) that encloses the ATP‐binding site. Within the cavity, several lysine residues are required for

Hydrogen‐Deuterium Exchange Mass Spectrometry for Molecular Glue Characterization

open access: yesAdvanced Science, EarlyView.
Molecular glues that stabilize protein–protein interactions are challenging to identify. Here, hydrogen‐deuterium exchange mass spectrometry can show and reveal gluing mechanisms that vary between protein binding partners. Abstract Molecular glues are powerful bioactive molecules that stabilize protein–protein interactions.

Metabolic Interplay in Acute Lung Injury: PARK7 Integrates FADS1/2‐Dependent PUFA Metabolism and H3K14 Lactylation to Attenuate Endothelial Ferroptosis and Dysfunction

open access: yesAdvanced Science, EarlyView.
SCAP/ARDS patients show reduced unsaturated fatty acids (UFAs) but higher PARK7 and lactate in plasma. PARK7 correlates positively with lactate and negatively with UFAs. Pulmonary endothelial cell multi‐omics show FADS1/2 downregulation and impaired PUFA metabolism. Lactate boosts PARK7 via H3K14 lactylation, and PARK7 then restores FADS1/2.

Procyanidin B3 and Its Derivatives Alleviate Neuronal Injury by Targeting G3BP1 for Ischemic Stroke Therapy

open access: yesAdvanced Science, EarlyView.
Ischemic stroke is a serious disease with high rates of mortality and disability, but there is a lack of novel therapeutic targets and agents for it. Now it is shown that Ras GTPase‐activating protein SH3 domain‐binding protein 1 (G3BP1) has the potential to serve as a therapeutic target for ischemic stroke.
