We screened acrylamide bioisosteres against KRasG12C and prioritized primary hits by their GSH reactivity, labeling efficiency and specificity. The most promising fragment was combined with a KRas‐binding scaffold. Covalent binding, inhibitory activity and selectivity were confirmed via MS, NMR, biochemical and cell viability assays.
