Potentiometric phosphate-sensing system utilizing phosphate-binding protein

Analytical and Bioanalytical Chemistry, 2002
A phosphate-detection system has been developed which uses phosphate-binding protein (PBP) from Escherichia coli. PBP was immobilized on a sheet of nitrocellulose membrane by cross-linking and the membrane potential of the immobilized PBP was measured. The response time of the system to phosphate was 5 min. The response was selective to phosphate among

The DING Family of Phosphate Binding Proteins in Inflammatory Diseases

2014
Human paraoxonase 1 (hPON-1) is a protein that has been studied in relation to its antioxidant and anti-atherosclerotic properties. Despite extensive studies, the molecular mechanisms responsible for its functional properties remain unclear. During the last decade, a new partner of hPON-1 has been identified. Hidden for a long time because of a similar

Mechanism of Pyridoxine 5′-Phosphate Accumulation in Pyridoxal 5′-Phosphate-Binding Protein Deficiency

Journal of Bacteriology, 2022
A PLP-binding protein (PLPBP) from the conserved COG0325 family has recently been recognized as a key player in vitamin B 6 homeostasis in various organisms. Loss of PLPBP disrupts vitamin B 6 homeostasis and perturbs diverse metabolisms, including amino acid and α-keto acid metabolism.

Tyrosine environment and phosphate binding in the archaebacterial histone-like protein HTa

Biochimica et Biophysica Acta (BBA) - Protein Structure and Molecular Enzymology, 1988
Abstract The thermophilic Archaebacterium, Thermoplasma acidophilum , has a DNA-stabilizing protein bound to its DNA. Each polypeptide chain of the protein contains only a single tyrosine residue. The protein exists normally as a complex of four identical subunits.

Phosphate-binding proteins and peptides: from molecular mechanisms to potential applications

Current Opinion in Biotechnology
Selective binding of phosphate is vital to multiple aims including phosphate transport into cells and phosphate-targeted applications such as adsorption-based water treatment and sensing. High-affinity phosphate-binding proteins and peptides offer a nature-inspired means of efficiently binding and separating phosphate from complex matrices. The binding

Phosphate binding protein as the biorecognition element in a biosensor for phosphate

Sensors and Actuators B: Chemical, 2004
This work explores the potential use of a member of the periplasmic family of binding proteins, the phosphate binding protein (PBP), as the biorecognition element in a sensing scheme for the detection of inorganic phosphate (Pi). The selectivity of this protein originates from its natural role which, in Escherichia coli, is to serve as the initial ...

A Biosensor for Inorganic Phosphate Using a Rhodamine-Labeled Phosphate Binding Protein

Biochemistry, 2006
A novel biosensor for inorganic phosphate (Pi) has been developed based on the phosphate binding protein of Escherichia coli. Two cysteine mutations were introduced and labeled with 6-iodoacetamidotetramethylrhodamine. When physically close to each other and correctly oriented, two rhodamine dyes interact to form a noncovalent dimer.

Phosphorin, A Phosphate-Binding Hydrophobic Protein Isolated from Renal Brush Border Membranes

1986
Reabsorption of inorganic phosphate (Pi) by the renal proximal tubule occurs from the tubular fluid across the brush border membrane into the cell interior by a Na+-dependent co-transport system. A number of the features of this Na+-Pi symport system have been documented by earlier workers(1,2). These include a description of the kinetic parameters and

Immunoblot detection of pyridoxal phosphate binding proteins in liver and hepatoma cytosolic extracts

Biochemical and Biophysical Research Communications, 1983
A monoclonal antibody, highly selective for the 5'-phosphopyridoxyl group, can be used to detect cytosolic pyridoxal-5'-phosphate binding proteins by an immunoblot procedure. This technique, when applied to sodium borohydride-treated cytosolic extracts obtained from normal rat liver at various stages of development as well as several liver-derived ...

Mechanism of Inorganic Phosphate Interaction with Phosphate Binding Protein fromEscherichia coli

Biochemistry, 1998
The mechanism of Pi interaction with phosphate binding protein of Escherichia coli has been investigated using the A197C mutant protein labeled with a coumarin fluorophore (MDCC-PBP), which gives a fluorescence change on binding Pi. A pure preparation of MDCC-PBP was obtained, in which the only significant inhomogeneity is the presence of equal amounts
