Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Inter-Limb Upper-Limb Strength Asymmetry and Rifle Shooting Performance Across Prone, Kneeling and Standing Positions in Elite Rifle Athletes

open access: yesApplied Sciences
Background: Rifle shooting performance depends on precise postural control, rifle stability, and coordinated upper-limb force production. Although previous studies have examined postural balance and aiming stability in rifle shooters, the role of upper ...

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Effect of Linear Sprints and Change-of-Direction Training Versus Small-Sided Soccer Games on Physical Performance in Highly Trained Young Female Soccer Players: A Randomized Cross-Over Study

open access: yesSports
Background: This study aimed to compare the effects of linear sprint training with changes of direction (LSCD) versus small-sided games (SSSG) on physical performance, agility, and soccer-specific skills in young elite female players.

Long‐term hippocampal alterations and cognitive impairment in a murine model of surgical sepsis

open access: yesFEBS Open Bio, EarlyView.
Using a mouse model of surgical sepsis, we tested long‐term memory and analyzed the transcriptome of single cells isolated from the hippocampus. Survivor mice showed worse memory, loss of certain brain cell subpopulations, and abnormal immune cell activity—suggesting that post‐sepsis brain alterations may be linked to cognitive deficits.

Molecular dynamics simulations of positively selected codons in FcγRI reveal novel biochemical binding properties

open access: yesFEBS Open Bio, EarlyView.
Evolutionary analysis across 32 placental mammals identified positive selection at residues H148 and W149 in the immune receptor FcγR1. Ancestral reconstruction combined with molecular dynamics simulations reveals how these mutations may influence receptor structure and dynamics, providing insight into the evolution of antibody recognition and immune ...

Versatile vector tools for efficient protein screening across multiple expression systems

open access: yesFEBS Open Bio, EarlyView.
A unified vector toolkit enables rapid protein expression screening across E. coli, insect, and mammalian cells. A single primer pair amplifies the target gene, which is inserted into any vector via a standardized interface. This streamlined workflow eliminates repeated cloning steps, accelerating the identification of optimal expression conditions for

PHYSICAL FORCE AS AN ADMISSIBILITY CONDITION IN PHYSICS

open access: yes
This paper isolates physical force as a candidate admissibility condition for physical reality and tests it under a strict elimination-by-necessity framework. Force is examined solely as a physically enforcing mechanism acting in spacetime, independent of mathematical formalisms, descriptions, or interpretations.
