Postsynthetic Modifications of Amide‐Enaminone‐Based [2]Rotaxanes for the Stereocontrolled Synthesis of Functionalized β‐Lactams

Novel amido‐enaminone threads efficiently template the formation of hydrogen‐bonded [2]rotaxanes and enable diverse postsynthetic modifications. Benzyl groups promote diastereoselective cyclizations to interlocked lactams while the enaminone motif allows stopper exchange while preserving the mechanical bond.

Studying the Outcomes and Mechanisms of Carbocationic Rearrangements Using Algorithm‐Augmented Experimentation

HopCat, a hybrid algorithm combining rule‐based transformations with quantum‐informed kinetics, can be applied to acid‐catalyzed terpenoid rearrangements. In synergy with human chemists and nuclear magnetic resonance analysis, it facilitates product assignment, candidate selection, and reconstruction of complex mechanistic pathways.

Remote Conformational Control of a Chloride‐Binding Site

Chloride‐binding foldamers have been developed that have stereogenic C‐ and N‐terminal groups separated by a helical α‐aminoisobutyric acid (Aib) body. Chloride affinity is sensitive to the sense and strength of screw‐sense induction by each N‐terminal residue.
