The Production of Polyhydroxybutyrate by Two-Step Fermentation and the Application of Polyhydroxybutyrate as a Novel Substrate for a Biolubricant

Two-step fermentation was utilized to increase PHB production. Cupriavidus necator was firstly cultivated in molasses medium for 144 h. Afterward, cells were recovered and extracted for PHB. The highest biomass (2.10 g/L) and PHB (62.38% of DCW) were obtained in the first reactor after 108 h of incubation.

Polyhydroxybutyrate (PHB) can be used as an alternative polymer to polylactide-glycolides for drug carrier production. It is a linear homopolymer biosynthesized by various strains of bacteria by condensation of D(-)-B-hydroxybutyric acid and used as an energy and carbon source.

The influence of a variety of treatments on the mobility and crystallinity of poly(hydroxybutyrate) (PHB) in whole cells and native granules has been proved using 13C-n.m.r. spectroscopy and X-ray powder diffraction, and correlated with the known biological effects of these treatments.
