ERRFI1, a neural crest (NC)‐associated gene, was upregulated in melanoma and negatively correlated with the expression of melanocytic differentiation markers and the susceptibility of melanoma cells toward BRAF inhibitors (BRAFi). Knocking down ERRFI1 significantly increased the sensitivity of melanoma cells to BRAFi.

Strength through diversity: how cancers thrive when clones cooperate
Intratumor heterogeneity can offer direct benefits to the tumor through cooperation between different clones. In this review, Kuiken et al. discuss existing evidence for clonal cooperativity to identify overarching principles, and highlight how novel technological developments could address remaining open questions.

Herds of Featherless Bipeds: Division and Privation in Plato’s Statesman
This paper explores privation in the Statesman, first from a methodological point of view, and then politically. I begin (§1) with the Rule of Bisection (262a-e): classes should be bifurcated by form, which apparently excludes division by positive and ...

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Spinoza and the problem of other substances [PDF]
Most of Spinoza’s arguments for God’s existence do not rely on any special feature of God, but instead on merely general features of substance. This raises the following worry: those arguments prove the existence of non-divine substances just as much as ...

LDAcoop: Integrating non‐linear population dynamics into the analysis of clonogenic growth in vitro
Limiting dilution assays (LDAs) quantify clonogenic growth by seeding serial dilutions of cells and scoring wells for colony formation. The fraction of negative wells is plotted against cells seeded and analyzed using the non‐linear modeling of LDAcoop.

Male muscle strength response to two methodologies for 1RM testing
The aims of this study were to valuate and to compare the weight moved during the execution of the 1 RM test using two different methodologies: with and without visual privation. The sample was composed of twelve men with age 29 ± 7.9 years, weight 79.1±
