Retroviruses incorporate themselves into a host genome in the form of a provirus, and can be passed on to the host's offspring. Evidence now emerges that one such provirus has been subverted to benefit of humans and other mammals that have a placenta — its protein product, called syncitin, may play an essential part in placental formation.

Integration of retroviral DNA appears to occur randomly in host genomes, suggesting that retroviruses can act as insertion mutagens. We have confirmed this prediction by showing that the nontransforming retrovirus, Moloney murine leukemia virus (M-MuLV), can insert its provirus within the selectable target provided by a single provirus in a clonal rat ...

Structure of a Defective Provirus of Bovine Leukemia Virus
Microbiology and Immunology, 1987AbstractDefective proviruses of bovine leukemia virus (BLV) in the genomes of infected cells were investigated by using Southern blotting hybridization analysis with various portions of a cloned BLV DNA as probes. When nine independent tumors of enzootic bovine leukosis with a single proviral copy per cell were examined, a single defective provirus of ...

A physical map of caprine arthritis-encephalitis provirus
Virology, 1984Terminally redundant linear proviral DNA of approximately 9.5 kb was the major unintegrated species recovered in the Hirt supernatant fraction of caprine synovial membrane cells infected with strain 75-G63 caprine arthritis-encephalitis virus. A physical map based on the cleavage sites of 13 restriction endonucleases was deduced for this proviral DNA.

I have discussed the observations and experiments that led to the formulation and establishment of the provirus hypothesis and the DNA provirus hypothesis, which includes RNA-directed DNA synthesis for the formation of the provirus. I have also discussed some aspects of the present status of our knowledge of the mechanism of formation of the ...
