Reductive Release from a Hybrid PKS‐NRPS during the Biosynthesis of Pyrichalasin H

The reductive release of cytochalasin intermediates was studied in vitro, showing that the R‐domain requires a T‐domain tethered substrate, but has low selectivity for the amino acid and polyketide moieties. Direct 2‐electron reduction is the preferred mode of release, followed by rapid Knoevenagel cyclisation of the released aldehyde.
