Identification of high-affinity pyridoxal kinase inhibitors targeting cancer therapy: an integrated docking and molecular dynamics simulation approach

Journal of Biomolecular Structure and Dynamics, 2023
Pyridoxal kinase (PDXK) is a vitamin B6-dependent transferase enzyme encoded by the PDXK gene, crucial for leukemic cell proliferation. Disruption of its activity causes altered metabolism and reduced levels of nucleotides and polyamines.

Thiamine‐dependent regulation of mammalian brain pyridoxal kinase in vitro and in vivo

Journal of Neurochemistry, 2022
Vitamins B1 (thiamine) and B6 (pyridox (al/ine/amine)) are crucial for central nervous system (CNS) function and neurogenesis due to the coenzyme action of their phosphorylated derivatives in the brain metabolism of glucose and neurotransmitters.

Pyridoxal Kinase

Annals of the New York Academy of Sciences, 1990
Chymotryptic digestion of sheep brain pyridoxal kinase, a dimer of identical subunits each of 40 kDa, yields two fragments of 24 and 16 kDa with concomitant loss of catalytic activity. These fragments were separated by HPLC and used for binding studies with ATP and pyridoxal analogues.

The interactions between biogenic amines and pyridoxal, pyridoxal phosphate, and pyridoxal kinase

Archives of Biochemistry and Biophysics, 1972
Abstract The effect of biogenic amines on pyridoxal kinase activity has been investigated in vitro . Experiments with a purified beef brain pyridoxal kinase indicate that 3,4-dihydroxyphenylalanine (dopa), dopamine, norepinephrine, and serotonin inhibit the reaction, while tyrosine, tryptophan, and 5-hydroxytryptophan (5-HTP) have no effect. However,

Crystal structure of human pyridoxal kinase

Journal of Structural Biology, 2006
Pyridoxal kinase, a member of the ribokinase superfamily, catalyzes the ATP-dependent phosphorylation reaction of vitamin B6 and is an essential enzyme in the formation of pyridoxal-5'-phosphate, a key cofactor for over 100 enzymes. Pyridoxal kinase is thus regarded as a potential target for pharmacological agents.

Interaction between Pyridoxal Kinase and Pyridoxal-5-phosphate-Dependent Enzymes

Journal of Biochemistry, 2003
The interactions of two pyridoxal-5-phosphate (PLP)-dependent enzymes, alanine aminotransferase (ALT) and glutamate decarboxylase (GAD), with pyridoxal kinase (PK) were studied by fluorescence polarization as well as surface plasmon resonance techniques. The results demonstrated that PK can specifically bind to ALT and GAD.

Pyridoxal kinase immunoreactivity in rabbit brain

Neurochemical Research, 1994
Murine polyclonal antibody against purified bovine brain pyridoxal kinase (EC 2.7.1.35) was generated and showed cross-reactivity with rabbit brain pyridoxal kinase. This antibody was used to immunohistochemically examine the distribution of pyridoxal kinase in the rabbit brain.
