Archives of Biochemistry and Biophysics, 2021Pyrroline-5-carboxylate reductase (PYCR in humans) catalyzes the final step of l-proline biosynthesis by catalyzing the reduction of L-Δ1-pyrroline-5-carboxylate (L-P5C) to l-proline using NAD(P)H as the hydride donor. In humans, three isoforms PYCR1, PYCR2, and PYCR3 are known.

Journal of Molecular Biology, 2006Pyrroline-5-carboxylate reductase (P5CR) is a universal housekeeping enzyme that catalyzes the reduction of Delta(1)-pyrroline-5-carboxylate (P5C) to proline using NAD(P)H as the cofactor. The enzymatic cycle between P5C and proline is very important for the regulation of amino acid metabolism, intracellular redox potential, and apoptosis.

Pyrroline-5-carboxylate reductase from Cucurbita cotyledons
Phytochemistry, 1973Pyrroline-5-carboxylate reductase, which required reduced pyridine nucleotide and Δ′-pyrroline-5-carboxylate for proline synthesis, was isolated from pumpkin cotyledons. The enzyme was found in the soluble fraction and had a 4.5-fold greater activity with NADH than NADPH. The enzyme was inhibited by NH2OH, NADP, ATP and slightly by proline. Glutathione

Analytical Biochemistry, 1973Abstract We describe a radioisotopic assay for Δ 1 -pyrroline-5-carboxylate reductase. In this assay we use Δ 1 -pyrroline-5-carboxylate[U- 14 C] and isolate product l -[U- 14 C]proline by cation-exchange column chromatography.
