Click chemistry–based proteomics revealed dynamic remodeling of the S‐palmitoylated proteome in dendritic cells upon TLR9 activation. Transcriptomics identified TLR9‐driven regulation of ZDHHC genes, including Zdhhc9. Genetic approaches uncovered potential ZDHHC9‐dependent targets.
