Peroxidasin enables melanoma immune escape by inhibiting natural killer cell cytotoxicity
Peroxidasin (PXDN) is secreted by melanoma cells and binds the NK cell receptor NKG2D, thereby suppressing NK cell activation and cytotoxicity. PXDN depletion restores NKG2D signaling and enables effective NK cell–mediated melanoma killing. These findings identify PXDN as a previously unrecognized immune evasion factor and a potential target to improve

Digital twins to accelerate target identification and drug development for immune‐mediated disorders
Digital twins integrate patient‐derived molecular and clinical data into personalised computational models that simulate disease mechanisms. They enable rapid identification and validation of therapeutic targets, prediction of drug responses, and prioritisation of candidate interventions.

Vignettes to Support Theory Refinement: Methodological Insights From a Realist Evaluation
Realist evaluation methodology aims to understand social programmes by revealing what works, for whom, in what circumstances, and how and why. Realist evaluation starts with generating initial programme theories (IPTs), which are subsequently tested and ...

Building Realist Program Theory for Large Complex and Messy Interventions
Program theory, that is, the specific idea about how a program causes the intended or observed outcomes, should be the central aspect of any realist evaluation or synthesis.

Objectives There is growing interest in evaluating new strategies to delay or prevent post‐traumatic osteoarthritis (PTOA) in individuals who have sustained anterior cruciate ligament (ACL) injury. This study sought to determine characteristics of potential treatments that are acceptable to patients with ACL injury.

Reflecting on Reflexivity in Realist Evaluation: A Call to Action
Realist evaluation is increasingly utilised across disciplines due to the value of identifying which mechanisms may explain how, and why, particular outcomes are generated in specific contexts.

Predicting extreme defects in additive manufacturing remains a key challenge limiting its structural reliability. This study proposes a statistical framework that integrates Extreme Value Theory with advanced process indicators to explore defect–process relationships and improve the estimation of critical defect sizes. The approach provides a basis for
