S100A8/A9‐high macrophages are markedly enriched in the stenotic intestinal tissue of patients with Crohn's disease. These profibrotic macrophages secrete mCCL6 in a STAT3‐dependent manner. mCCL6 and its human ortholog hCCL15 activate fibroblasts via the CCR1 receptor, thereby driving excessive collagen deposition.

Astrocytic FABP5 promotes mitochondrial stress, cGAS‐STING pathway activation, pyroptosis, and neuroinflammation in epilepsy, contributing to seizure pathology. Genetic targeting of FABP5 or pharmacological inhibition of STING alleviates epileptic phenotypes, highlighting a potential therapeutic strategy for epilepsy.

In sepsis‐induced liver injury, marked neutrophil infiltration correlates with disease severity. Neutrophil SLC11A1 drives intracellular Fe2+ and ROS via the Fenton reaction, promoting NETs formation. Hepatocyte‐derived CXCL10 recruits neutrophils and activates the JAK/STAT1/SLC11A1 axis, enhancing NETs‐mediated inflammation.

Identification of candidate substrates for ectodomain shedding by the metalloprotease-disintegrin ADAM8 [PDF]
ADAM proteases are type I transmembrane proteins with extracellular metalloprotease domains. As for most ADAM family members, ADAM8 (CD156a, MS2) is involved in ectodomain shedding of membrane proteins and is linked to inflammation and neurodegeneration.
