Molecular, spatial, and AI‐enabled technologies for predicting neoadjuvant therapy response in locally advanced rectal cancer

open access: yesThe Journal of Pathology, EarlyView.
Abstract Pretreatment prediction of neoadjuvant response remains an unresolved challenge in locally advanced rectal cancer, where management is moving towards risk‐adapted and response‐oriented treatment strategies. Prediction remains difficult because response reflects interacting tumour‐intrinsic, immune, stromal, and spatial processes that are ...

Feasibility of Urethra‐Sparing Ultra‐Hypofractionated Three‐Dose‐Level Radiotherapy for Prostate Cancer with a Dominant Intraprostatic Lesion Abutting the High‐Dose Avoidance Zone

open access: yesPrecision Radiation Oncology, EarlyView.
Ultra‐hypofractionated three‐dose‐level radiotherapy allows safe dose escalation to dominant intraprostatic lesions near the urethra while maintaining full coverage of pelvis and prostate and respecting urethral and organ‐at‐risk constraints. Abstract This planning study evaluated ultra‐hypofractionated three‐dose‐level radiotherapy for prostate cancer

Evaluation of Concordance between Radiation Therapists and Medical Physicists in MR‐Guided Online Adaptive Planning

open access: yesPrecision Radiation Oncology, EarlyView.
Magnetic resonance (MR)‐guided online adaptive radiotherapy requires skill in online adaptive treatment planning. This study demonstrated that radiation therapists, with appropriate training, can consistently produce high‐quality adaptive plans concordant with those created by medical physicists for treatment on a 1.5 Tesla MR‐Linac.

Exploratory Analysis of Peri‐Prostatic Adipose Tissue Radiomics Features and Their Potential to Predict Prostate Cancer Diagnosis: Results From a UK Biobank Study

open access: yesThe Prostate, EarlyView.
ABSTRACT Introduction This paper reports the development and application of an artificial intelligence (AI)–led approach to large volume MRI data curation and image segmentation of periprostatic adipose tissue (PPAT) in prostate cancer (PCa)–free men from the UK Biobank. We report how PPAT was defined, radiomics features of PPAT extracted, and explored

Comparison of Gene Enrichment Between Prostate Tissue and Urine for Non‐Invasive Biomarker Development

open access: yesThe Prostate, EarlyView.
ABSTRACT Background For prostate cancer (PCa), early diagnosis and biomarker‐based patient stratification can improve clinical decision‐making and post‐treatment prognosis. PCa biomarkers are best validated in tissue following comprehensive histopathologic review; however, urinary biomarkers can be obtained non‐invasively and are therefore safer and ...

Urinary Soluble Collagen Levels Are Associated With Moderate/Severe LUTS

open access: yesThe Prostate, EarlyView.
ABSTRACT Background Expansion of the stromal compartment of the prostate concurrent with fibroblast activation and increased collagenization is now recognized as a pathobiology likely contributing to Lower Urinary Tract Symptoms (LUTS). However, approaches to quantitatively and, ideally, non‐invasively, assess prostatic collagenization in vivo are not ...

Circulating Tumor Cell Synaptophysin and Androgen Receptor Expression Reflect Prostate Cancer Phenotypes in Metastatic Disease

open access: yesThe Prostate, EarlyView.
ABSTRACT Background Chronic exposure to androgen receptor (AR)‐targeted therapies can drive lineage plasticity, leading to treatment resistance and neuroendocrine prostate cancer (NEPC), characterized by loss of AR expression and acquisition of neuroendocrine (NE) markers including synaptophysin (SYP). We evaluated concordance between circulating tumor
