Diversity and complexity in neural organoids

open access: yesFEBS Letters, EarlyView.
Neural organoid research aims to expand genetic diversity on one side and increase tissue complexity on the other. Chimeroids integrate multiple donor genomes within single organoids. Self‐organising multi‐identity organoids, exogenous cell seeding, or enforced assembly of region‐specific organoids contribute to tissue complexity.

Modulation of Homer1 EVH1 domain internal dynamics by putative autism‐associated mutations

open access: yesFEBS Letters, EarlyView.
The putative autism‐associated M65I and S97L variants of the EVH1 domain of the postsynaptic scaffold protein Homer1 do not exhibit substantial changes in their overall structure or partner binding. Both of them, but especially the M65I variant, show altered internal dynamics relative to the wild‐type domain on the μs‐ms timescale, indicated by the ...

Stratification Theory and regularity conditions

open access: yes, 2007
Na presente dissertação faremos um estudo dos conjuntos algébricos, semialgébricos, analíticos, semianalíticos e subanalíticos, real e complexo, através das condições de regularidade da estratificação destes conjuntos.

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Heterozygous loss‐of‐function alleles associate the conserved 3′‐5′ exoribonuclease EXOSC10 with hypersensitivity to the anticancer drug 5‐fluorouracil

open access: yesMolecular Oncology, EarlyView.
EXOSC10, an essential nuclear RNA exosome‐associated 3′‐5′ exoribonuclease, is inhibited by the anticancer drug 5‐fluorouracil (5‐FU), and EXOSC10 depletion increases 5‐FU sensitivity. The colon‐cancer variant EXOSC10S402T, located in a proteolysis motif, is stable and nuclear but nonfunctional in vivo.

Regularity for 3D Navier-Stokes equations in terms of two components of the vorticity

open access: yesElectronic Journal of Differential Equations, 2010
We establish regularity conditions for the 3D Navier-Stokes equation via two components of the vorticity vector. It is known that if a Leray-Hopf weak solution $u$ satisfies $$ ilde{omega}in L^{2/(2-r)}(0,T;L^{3/r}(mathbb{R}^3))quad hbox{with }0< ...

Cell‐cycle‐specific lesion evolution rather than inhibition of double‐strand‐break repair underpins cisplatin radiosensitization

open access: yesMolecular Oncology, EarlyView.
We analyze cisplatin–DNA adducts (CDAs) and double‐strand breaks (DSBs) in a cell‐cycle‐dependent manner. We find that CDAs form similarly across all cell cycle phases. DSBs arise only in S‐phase. CDAs might not directly impair DSB repair, but S‐phase DSB lesions evolve in the presence of CDAs and disrupt repair in G2, also causing radiosensitization ...

Calculation of stationary points in linear bilevel programming problems

open access: yesДоклады Белорусского государственного университета информатики и радиоэлектроники, 2019
Bilevel programming problems are considerably difficult for numerical analysis. Despite the vast amount of literature and research dedicated to studying them, there are no universal methods to solve them and numerous publications are concerned mainly ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Elliptic and parabolic regularity for second‐ order divergence operators with mixed boundary conditions

open access: yes, 2016
We study second-order equations and systems on non-Lipschitz domains including mixed boundary conditions. The key result is interpolation for suitable function spaces. From this, elliptic and parabolic regularity results are deduced by means of Šneı̆berg'
