Targeting Supramolecular Active Complexes of Nav1.7/Nav1.8 to Relieve Chronic Neuropathic Pain
In mice and patients with severe chronic neuropathic pain (NP), Nav1.7, Nav1.8, TrkB, and five cytoskeletal proteins form supramolecular active complexes (SMACs) with polygonal lattice structures as noxious signal amplifiers in dorsal root ganglion (DRG) neurons.

METTL3‐mediated m6A modification stabilizes KDR/Kdr mRNA through YTHDC2, amplifying the VEGFA‐KDR feedback loop and disturbing Th17/Treg balance. This pathway promotes persistent inflammation and tissue damage in patients with AIT, and inhibiting the METTL3‐KDR axis effectively disrupts this circuit, alleviating thyroid tissue injury and disease ...

DDX3 depletion selectively represses translation of structured mRNAs
Abstract DDX3 is an RNA chaperone of the DEAD-box family that regulates translation. Its yeast ortholog Ded1 controls the translation of nearly all mRNAs, whereas DDX3 is thought to regulate only a subset of mRNAs. However, the set of mRNAs that are regulated by DDX3 are unknown, along with the relationship between DDX3 binding and activity.

In this article, Shuai and colleagues demonstrate that metabolic remodeling drives self‐diploidization in murine haploid ESCs (haESCs). Mitochondrial dysfunction and imbalanced pyruvate metabolism underlie this process. Genome‐wide screening using haESCs identifies key mitochondrial quality‐control related genes, enabling a metabolism‐based medium that

ANKS1B in the nucleus accumbens plays a critical role in the transition from controlled to escalated cocaine intake. Mechanistically, ANKS1B interacts with CBP to epigenetically suppress FoxO3 through H3K27 acetylation. The ANKS1B‐CBP‐FoxO3 signaling cascade presents a novel theraputic target for the treatment of cocaine addiction.

The BnTFL1‐BnJAM3‐BnSWEETs Module Orchestrates Seed Storage Reserve Accumulation in Brassica napus
BnaC03.TFL1 and BnaA01.JAM3 form a complex that facilitates the transport of soluble sugars from the seed coat to the embryo by directly activating the expression of BnSWEETs. Furthermore, BnaC03.TFL1 and BnaA01.JAM3 work cooperatively to promote soluble sugar and oil production while suppressing protein deposition.

Yan polymerization contributes to active repression and thus contributes to precise gene expression
Downstream of the receptor tyrosine kinase (RTK) pathway in Drosophila, a conserved network mediates the robust regulation of genes involved in developmentally important processes such as cell proliferation and differentiation.
