METTL10‐Mediated PIAS3 Methylation Links Purine Metabolism to Gastric Cancer Progression

open access: yesAdvanced Science, EarlyView.
METTL10‐mediated methylation of PIAS3 at K442 inhibits its SUMO E3 ligase activity toward MITF. This reduces MITF SUMOylation and subsequent RNF4‐mediated ubiquitination, leading to MITF protein accumulation and gastric cancer progression. Abstract Metabolic dysregulation plays a significant role in the development of gastric cancer (GC).

Deciphering Neural Mechanisms Underlying Marmoset Dynamic Natural Behaviors Using a Miniaturized Wireless Large‐Scale Coverage Neural Recorder

open access: yesAdvanced Science, EarlyView.
This study introduces a miniaturized wireless neural recording system integrated with custom‐designed 120‐channel µECoG arrays, engineered for monitoring brain activities in freely behaving marmosets. The system can capture and characterize behavior‐specific neural signals, including drinking‐related activation, pre‐vocalization anticipatory responses,

Engineered crRNA Drives RPA‐T7‐CRISPR/Cas14a Cascade for Ultrasensitive Detection of ctDNA PIK3CA H1047R

open access: yesAdvanced Science, EarlyView.
The TIDE‐Cas14a system, an integrated RPA‐T7‐CRISPR/Cas14a cascade platform incorporating engineered crRNA mismatches, enables ultrasensitive detection of the PIK3CA H1047R mutation in breast cancer circulating tumor DNA, with a limit of detection as low as 0.01%, surpassing the sensitivity of droplet digital PCR.
