Targeting ROCK2 to Restore Epileptic Synaptic Networks via Mitophagy Activation: Insights from Translational Imaging of SV2A In Vivo

open access: yesAdvanced Science, EarlyView.
By integrating synaptic vesicle glycoprotein 2A PET imaging and transcriptomics, this study identifies Rho‐associated protein kinase 2 (ROCK2) as a critical regulator of synaptic network dysfunction in temporal lobe epilepsy. Pharmacological ROCK2 inhibition restores synaptic density and enhances mitophagy, thereby reducing seizures and neuronal damage.

Interpretable Multimodal Fusion Model Enhances Postoperative Recurrence Prediction in Gastric Cancer

open access: yesAdvanced Science, EarlyView.
An interpretable multimodal fusion model (RSA) integrating clinical, radiomic, and pathomic features is developed and validated to predict early postoperative recurrence in gastric cancer. The RSA model demonstrates superior predictive performance and holds potential for guiding adjuvant chemotherapy decisions and improving individualized patient ...

Microphysiological Solid Tumor Models in Hydrogel Beads for CAR T Cell Immunotherapy Evaluation

open access: yesAdvanced Science, EarlyView.
The complex physical and biochemical barriers of the tumor microenvironment are major triggers of immune exclusion. Here, a 3D micrometastasis model is developed to mimic key microenvironmental cues, enabling in vitro evaluation of chimeric antigen receptor (CAR) T cell immunotherapy. Abstract Micrometastases are challenging to resect surgically and to

SCORCH2: A Generalized Heterogeneous Consensus Model for High‐Enrichment Interaction‐Based Virtual Screening

open access: yesAdvanced Science, EarlyView.
Researchers developed SCORCH2, an advanced machine‐learning scoring function for virtual screening that combines two complementary XGBoost models to re‐evaluate protein‐ligand binding plausibility. The method demonstrates superior performance on standard benchmarks, shows excellent generalization to previously unseen protein targets, and provides ...
