Investigation of Squaramide Catalysts in the Aldol Reaction En Route to Funapide

The asymmetric aldol reaction of an industrially viable synthesis of funapide, a promising analgesic agent, was studied with a range of squaramide catalysts. Compared to previously employed thiourea structures, these catalysts provided a non‐negligible improvement in the enantioselectivity of the reaction, while keeping similar activity.

A new sustainable method for synthesizing 1,3‐N,S‐heterocycles, including 1,3‐thiazolidine‐2‐imines and 4,5‐dihydrothiazol‐2‐amines is reported. This innovative approach offers considerable shorter reaction times with higher yields, reduced energy consumption, and the elimination of hazardous chemicals.
