Spatiotemporal and quantitative analyses of phosphoinositides – fluorescent probe—and mass spectrometry‐based approaches

open access: yesFEBS Letters, EarlyView.
Fluorescent probes allow dynamic visualization of phosphoinositides in living cells (left), whereas mass spectrometry provides high‐sensitivity, isomer‐resolved quantitation (right). Their synergistic use captures complementary aspects of lipid signaling. This review illustrates how these approaches reveal the spatiotemporal regulation and quantitative

The anti‐CRISPR protein AcrIE8.1 inhibits the type I‐E CRISPR‐Cas system by directly binding to the Cascade subunit Cas11

open access: yesFEBS Letters, EarlyView.
In this study, we present the structure of AcrIE8.1, a previously uncharacterized anti‐CRISPR protein that inhibits the type I‐E CRISPR‐Cas system. Through a combination of structural and biochemical analyses, we demonstrate that AcrIE8.1 directly binds to the Cas11 subunit of the Cascade complex to inhibit the CRISPR‐Cas system.

An automated method for high-throughput protein purification applied to a comparison of His-tag and GST-tag affinity chromatography

open access: yesBMC Biotechnology, 2003
Background Functional Genomics, the systematic characterisation of the functions of an organism's genes, includes the study of the gene products, the proteins. Such studies require methods to express and purify these proteins in a parallel, time and cost

Transitions between homophilic and heterophilic modes of cooperation

open access: yes, 2015
Cooperation is ubiquitous in biological and social systems. Previous studies revealed that a preference toward similar appearance promotes cooperation, a phenomenon called tag-mediated cooperation or communitarian cooperation.

An intracellular transporter mitigates the CO2‐induced decline in iron content in Arabidopsis shoots

open access: yesFEBS Letters, EarlyView.
This study identifies a gene encoding a transmembrane protein, MIC, which contributes to the reduction of shoot Fe content observed in plants under elevated CO2. MIC is a putative Fe transporter localized to the Golgi and endosomal compartments. Its post‐translational regulation in roots may represent a potential target for improving plant nutrition ...

Distributional, differential and integral problems: equivalence and existence results

open access: yesElectronic Journal of Qualitative Theory of Differential Equations, 2017
We are interested in studying the matter of equivalence of the following problems: \[ \begin{aligned} Dx&=f(t,x)Dg\nonumber \\ x(0)&=x_0 \end{aligned}\tag{1} \label{1a} \] where $Dx$ and $Dg$ stand for the distributional derivatives of $x$ and $g ...

A Cre‐dependent lentiviral vector for neuron subtype‐specific expression of large proteins

open access: yesFEBS Letters, EarlyView.
We designed a versatile and modular lentivector comprising a Cre‐dependent switch and self‐cleaving 2A peptide and tested it for co‐expression of GFP and a 2.8 kb gene of interest (GOI) in mouse cortical parvalbumin (PV+) interneurons and midbrain dopamine (TH+) neurons.

An ER Protein Functionally Couples Neutral Lipid Metabolism on Lipid Droplets to Membrane Lipid Synthesis in the ER

open access: yesCell Reports, 2014
Eukaryotic cells store neutral lipids such as triacylglycerol (TAG) in lipid droplets (LDs). Here, we have addressed how LDs are functionally linked to the endoplasmic reticulum (ER). We show that, in S. cerevisiae, LD growth is sustained by LD-localized
