Cloning of a cDNA encoding the Saussurea medusa chalcone isomerase and its expression in transgenic tobacco

Chalcone isomerase (CHI; EC 5.5.1.6) is a key enzyme in the flavonoid biosynthesis pathway. We isolated a CHI gene (SmCHI) from a cDNA library derived from Saussurea medusa (Asteraceae) cell cultures. The cDNA and genomic sequences of SmCHI are the same; in other words, this gene is intronless.

[Cloning, expression and charaterization of chalcone synthase from Saussurea medusa].

A fragment of chalcone synthase gene (SmCHS) was cloned from the cDNA library constructed in Saussurea medusa. The full-length cDNA sequence of SmCHS was obtained by RT-PCR. Sequence analysis showed that the full length of SmCHS was 1313 bp, containing an open reading frame (1170 bp) encoding 389 amino acids.

Cellular aggregate size as the critical factor for flavonoid production by suspension cultures of Saussurea medusa

Three previously established cell lines (yellow, red and white) of Saussurea medusa were investigated for jaceosidin and hispidulin production. Maximum yields of the jaceosidin and hispidulin were obtained in the red cell line at 75+/-0.41 and 6.4+/-0.31 mg l-1.

Optimization of growth and jaceosidin production in callus and cell suspension cultures of Saussurea medusa

Calli cultures derived from the leaves of Saussurea medusa were selected on the basis of colour into three callus, A, B and C, which suggested different levels of metabolite accumulation. An improved reversed phase high performance liquid chromatographic method provided selective determination of the jaceosidin content of these samples.

Abstract Suspension cultures of Sussurea medusa produce jaceosidin and hispidulin. In an attempt to increase productivity, silver nitrate and glutathione (GSH) elicitors were tested. Both dosage and addition time as well as time course studies were carried out on the effects of the elicitors on the formation of the metabolites.
