Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Beyond Scarcity: Perspectives on Energy Transition

open access: yesRelations, 2018
Two dominant lines of reasoning in the philosophical debate on energy transition can be described as boundless consumerism (we should find ways to keep growing) and eco-frugality (we should reduce our impact as much as possible). This paper problematizes

On simplicity and emergence [PDF]

open access: yesPsychonomic Bulletin & Review, 2017
Mark Johnson's paper centres around a provocative and sensible point: the simplicity of a change in evolutionary terms does not necessarily map straightforwardly onto the simplicity of that change within aformal system. This commentary discusses these implications in more depth, making the point that it isimportant to consider the level at which an ...

Heterozygous loss‐of‐function alleles associate the conserved 3′‐5′ exoribonuclease EXOSC10 with hypersensitivity to the anticancer drug 5‐fluorouracil

open access: yesMolecular Oncology, EarlyView.
EXOSC10, an essential nuclear RNA exosome‐associated 3′‐5′ exoribonuclease, is inhibited by the anticancer drug 5‐fluorouracil (5‐FU), and EXOSC10 depletion increases 5‐FU sensitivity. The colon‐cancer variant EXOSC10S402T, located in a proteolysis motif, is stable and nuclear but nonfunctional in vivo.

Simplicity and stability of the first eigenvalue of a (p;q) Laplacian system

open access: yesElectronic Journal of Differential Equations, 2012
This article concerns special properties of the principal eigenvalue of a nonlinear elliptic system with Dirichlet boundary conditions. In particular, we show the simplicity of the first eigenvalue of $$displaylines{ -Delta_p u = lambda |u|^{alpha-1}

Perceptual Simplicity: The True Role of Prägnanz and Occam

open access: yesGestalt Theory, 2019
In recent years, the concept of simplicity in perception has acquired a leading role, above all thanks to scholars linked to Bayesian modeling and to theories like structural information theory derived from information theory.

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

COMPLEXITY * SIMPLICITY * SIMPLEXITY [PDF]

open access: yes
“In the midst of order, there is chaos; but in the midst of chaos, there is order”, John Gribbin wrote in his book Deep Simplicity (p.76). In this dialectical spirit, we discuss the generative tension between complexity and simplicity in the theory and ...
