A central role for STAT5 in the transcriptional programing of T helper cell metabolism
Activated lymphocytes adapt their metabolism to meet the energetic and biosynthetic demands imposed by rapid growth and proliferation. Common gamma chain (cγ) family cytokines are central to these processes, but the role of downstream signal transducer ...

Miz1 is a zinc finger transcription factor with an N-terminal POZ domain. Complexes with Myc, Bcl-6 or Gfi-1 repress expression of genes like Cdkn2b (p15(Ink4)) or Cdkn1a (p21(Cip1)).

We show that the majority of the 18 analyzed recurrent cancer‐associated ERBB4 mutations are transforming. The most potent mutations are activating, co‐operate with other ERBB receptors, and are sensitive to pan‐ERBB inhibitors. Activating ERBB4 mutations also promote therapy resistance in EGFR‐mutant lung cancer.

Subtype‐specific enhancer RNAs define transcriptional regulators and prognosis in breast cancers
This study employed machine learning methodologies to perform the subtype‐specific classification of RNA‐seq data sets, which are mapped on enhancers from TCGA‐derived breast cancer patients. Their integration with gene expression (referred to as ProxCReAM eRNAs) and chromatin accessibility profiles has the potential to identify lineage‐specific and ...

Tumors contain diverse cellular states whose behavior is shaped by context‐dependent gene coordination. By comparing gene–gene relationships across biological contexts, we identify adaptive transcriptional modules that reorganize into distinct vulnerability axes.

The biomimetic nanoplatform IMNP (ETS1 pDNA/PBAE@ITP‐MM) undergoes targeted disassembly at inflammatory vascular sites to release the ETS1 plasmid (pETS1). This release initiates a cascade of effects that inhibit pathogenic pathways and support immune homeostasis. (Abbreviations: EndMT, endothelial‐to‐mesenchymal transition; EC, endothelial cell; TC, T

The transcription factor STAT5 drives mutation and imatinib resistance in chronic myeloid leukemia via ROS production [PDF]
Background Chronic myelogenous leukemia (CML) is a leukemic stem cell (LSC)-driven myeloproliferative disorder and is associated with a characteristic chromosomal translocation which generates a constitutively active tyrosine kinase, the BCR-ABL oncoprotein.

IL‐15‐engineered stem cell–NK cell complexes, assembled via bioorthogonal chemistry, enable effective lung cancer immunotherapy. Abstract Natural killer (NK) cells represent a powerful immunotherapeutic strategy due to their intrinsic cytotoxicity and ability to target tumor cells independently of antigen presentation.
