CDK11 inhibition stabilises the tumour suppressor p53 and triggers the production of an alternative p21WAF1 splice variant p21L, through the inactivation of the spliceosomal protein SF3B1. Unlike the canonical p21WAF1 protein, p21L is localised in the cytoplasm and has reduced cell cycle‐blocking activity.

We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.

Stepped wedge cluster randomized controlled trials: sample size and power determinations [PDF]
Stepped wedge cluster randomized controlled trials (RCTs) are increasingly used in evaluating a causal-effect relationship between an intervention and an outcome. Sample size and power calculations are critical in designing a statistical study. Thus, the

Stepped wedge cluster randomized controlled trial designs: a review of reporting quality and design features [PDF]
Background The stepped wedge (SW) cluster randomized controlled trial (CRCT) design is being used with increasing frequency. However, there is limited published research on the quality of reporting of SW-CRCTs.

Comparative evaluation of the stepped wedge design efficiency [PDF]
The cluster randomised trial (CRT) design is widely used to evaluate interventions targeted at the cluster level and helps alleviate contamination within clusters. CRTs usually require large sample sizes, although matched pair CRTs can limit this problem.

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

Highly efficient stepped wedge designs for clusters of unequal size [PDF]
AbstractThe stepped wedge design (SWD) is a form of cluster randomized trial, usually comparing two treatments, which is divided into time periods and sequences, with clusters allocated to sequences. Typically all sequences start with the standard treatment and end with the new treatment, with the change happening at different times in the different ...
