This work represents a second observation, and the first crystallographic evidence, of true intercalation of a small‐molecule ligand (here, bisquinolinium PyDH2) into an antiparallel G‐quadruplex (G4) DNA fold. Abstract G‐quadruplexes (G4s) are non‐canonical DNA structures implicated in a number of biological processes.

Mitigating Molecular Aggregation in Drug Discovery With Predictive Insights From Explainable AI

In this work, MEGAN, our explainable AI model, is applied to predict small colloidally aggregating molecules (SCAMs), a significant source of false positives in drug discovery. MEGAN provides interpretable predictions and proposes structurally similar, non‐aggregating alternative molecules.

Structural Insight Into a Human H Ferritin@Gold‐Monocarbene Adduct: Aurophilicity Revealed in a Biological Context

Human H ferritin (HuHf) is a nanocarrier for anticancer metal drugs. By cryo‐EM, a 1.51 Å structure of its adduct with Au(NHC)Cl was solved, showing a novel tetranuclear gold(I) cluster bound to Cys90 and Cys102. Short inter‐metal distances indicate aurophilic interactions.

Protein Secondary Structure Patterns in Short‐Range Cross‐Link Atlas

Short‐range cross‐linking mass spectrometry (XL‐MS) reveals secondary structure‐dependent patterns in proteome‐wide datasets. By integrating XL‐MS data with AlphaFold‐predicted structures, we introduce X‐SPAN, a novel validation metric that assesses data quality and enables pLDDT score benchmarking.
