Retinoic Acid Reprograms Mast Cells Toward a Proinflammatory State to Enhance Antitumor Immunity

Proinflammatory mast cells represent an MHC‐IIhigh, cytokine‐producing mast cell subset associated with improved survival and enhanced responses to anti‐PD‐1 therapy. Retinoic acid drives their polarization, enabling antigen uptake and presentation, and T cell recruitment and activation, collectively promoting adaptive anti‐tumor immunity.

Sonoepigenetic Modification Mechanoprimes Early Osteogenic Commitment in Mesenchymal Stem Cells

Brief 10 min bursts of high frequency (10 MHz) vibrational mechanostimulation to stem cells are shown to activate their epigenetic machinery to condition their differentiation down an osteogenic lineage within just 3 days, which is much earlier than that with other forms of mechanostimuli, and without requiring any biochemical additives. Mechanical stiffening of the extracellular matrix around mature mammary acini induces sequential remodeling phases. An early response disrupts the basement membrane and promotes fibronectin secretion, followed by delayed invasion with matrix reorganization and proliferation.

Aloe‐Emodin Targeting FOXC2 Disrupts NETs Formation and EMT‐Driven Postoperative Peritoneal Adhesion Through TGF‐β1‐Smad2/3 Pathway

Forkhead box protein C2 (FOXC2) is highlighted as a pivotal effector in postoperative peritoneal adhesion (PPA) formation. Mechanically, FOXC2 mediates NETs‐induced EMT alterations through the TGF‐β1‐Smad2/3 pathway. Aloe‐emodin (AE) ameliorates FOXC2‐driven fibrosis by blocking NETs formation and EMT changes through this pathway.

ETV1 Drives CD4+ T Cell‐Mediated Intestinal Inflammation in Inflammatory Bowel Disease Through Amino Acid Transporter Slc7a5

This study identifies the transcription factor ETV1 as a key driver of CD4⁺ T cell‐mediated intestinal inflammation in inflammatory bowel disease (IBD). ETV1 promotes CD4⁺ T cell activation, proliferation, and Th17 differentiation by activating the amino acid transporter SLC7A5, fueling metabolic reprogramming.
