Shear Conditioning Promotes Microvascular Endothelial Barrier Resilience in a Human BBB‐on‐a‐Chip Model of Systemic Inflammation Leading to Astrogliosis

Physiological flow (0.5 Pa) conditioning promotes resilience of an iPSC‐derived blood–brain barrier endothelium to mild systemic inflammation. In contrast, high “cytokine storm” levels induce barrier breakdown, fibrinogen leakage, and astrocyte activation, reconstituting the two‐hit injury mechanism on‐a‐chip.

Therapy‐Induced ECM Remodeling Creates a Transient Immune Barrier in Residual Melanoma

BRAF/MEK inhibition promotes extracellular matrix (ECM) accumulation and remodeling during therapy‐tolerant residual disease, limiting CD8⁺ T cell access to tumor cells. Targeting ECM accumulation restores CD8⁺ T cell infiltration, enhances their intratumoral distribution, and delays therapeutic resistance.

Smohaze‐Upregulated RFWD3 Competes with TRIM24 to Stabilize TREX1 and Reduce Cytosolic dsDNA in Non‐Small Cell Lung Cancer

The E3 ubiquitin ligase RFWD3 binds the exonuclease TREX1 and protects it from TRIM24‐mediated ubiquitination and degradation, leading to the eradication of intracellular dsDNA, inhibition of the cGAS‐STING pathway, and immunosuppressive microenvironment of lung cancer.
