Chloride binding site of neurotransmitter sodium symporters [PDF]

open access: yesProceedings of the National Academy of Sciences, 2013
Neurotransmitter:sodium symporters (NSSs) play a critical role in signaling by reuptake of neurotransmitters. Eukaryotic NSSs are chloride-dependent, whereas prokaryotic NSS homologs like LeuT are chloride-independent but contain an acidic residue (Glu290 in LeuT) at a site where eukaryotic NSSs have a serine.

Iodine status in Turkish populations and exposure to iodide uptake inhibitors. [PDF]

open access: yesPLoS ONE, 2014
Perchlorate, nitrate, and thiocyanate are competitive inhibitors of the sodium iodide symporter of the thyroid membrane. These inhibitors can decrease iodine uptake by the symporter into the thyroid gland and may disrupt thyroid function.

Structure of eukaryotic purine/H+ symporter UapA suggests a role for homodimerization in transport activity

open access: yesNature Communications, 2016
UapA is a uric acid/xanthine H+symporter from a filamentous fungus. Here, the authors solve the crystal structure of the transporter in complex with xanthine revealing it to be a dimer, and this homodimerisation is proposed to be important for function.

General Principles of Secondary Active Transporter Function [PDF]

open access: yes, 2019
Transport of ions and small molecules across the cell membrane against electrochemical gradients is catalyzed by integral membrane proteins that use a source of free energy to drive the energetically uphill flux of the transported substrate. Secondary active transporters couple the spontaneous influx of a "driving" ion such as Na$^+$ or H$^+$ to the ...

Bilirubin Targeting WNK1 to Alleviate NLRP3‐Mediated Neuroinflammation

open access: yesAdvanced Science, EarlyView.
At physiological concentrations, bilirubin binds to the kinase domain of WNK1, thereby augmenting its activity and facilitating the phosphorylation of downstream SPAK/OSR1. This phosphorylation inhibits KCC2 activity, leading to elevate intracellular chloride levels in neurons.

Tumor microenvironment affects exogenous sodium/iodide symporter expression

open access: yesTranslational Oncology, 2021
For decades, sodium/iodide symporter NIS-mediated iodide uptake has played a crucial role in the radioactive ablation of thyroid cancer cells. NIS-based gene therapy has also become a promising tool for the treatment of tumors of extrathyroidal origin ...

Utilizing Caenorhabditis Elegans as a Rapid and Precise Model for Assessing Amphetamine‐Type Stimulants: A Novel Approach to Evaluating New Psychoactive Substances Activity and Mechanisms

open access: yesAdvanced Science, EarlyView.
This study uses Caenorhabditis elegans to assess the activity of amphetamine‐type stimulants (ATS) and their analogs. The research highlights the roles of dopamine and serotonin pathways, distinguishes chiral forms of ATS, and identifies structure‐activity relationships. These findings provide a rapid and cost‐effective model for evaluating the effects

Clinically compliant spatial and temporal imaging of chimeric antigen receptor T-cells

open access: yesNature Communications, 2018
Adoptive transfer of chimeric antigen receptor (CAR) T cells has shown promising anticancer results in clinical trials. Here the authors use the human sodium iodide symporter (hNIS) as a reporter gene to image human CAR T cells in cancer-bearing mice ...
