Environmental DNA, Volume 8, Issue 1, January–February 2026.This study used environmental DNA (eDNA) metabarcoding to assess vertebrate biodiversity in headwater streams of the Rio Santo Antônio basin, southeastern Brazil, a tributary of the Rio Doce. A total of 119 vertebrate OTUs were identified, with oxidation–reduction potential emerging as the strongest environmental predictor of species richness.
