A cellular system to study responses to a collision between the transcription complex and a protein‐bound nick in the DNA template

open access: yesFEBS Letters, EarlyView.
We present the cellular transcription‐coupled Flp‐nick system allowing the introduction of a Top1‐mimicking cleavage complex (Flpcc) at a Flp recognition target site within a controllable LacZ gene. LacZ transcription leads to the collision of RNA polymerase II (RNAPII) with Flpcc, and this causes RNAPII stalling, ubiquitination, and degradation.

Adaptation of a University Soils Mechanics Course for a Union Technician Training Program [PDF]

open access: yes, 2019
When dealing with the term “geotechnical”, most often engineers associate this with the engineering during the design phase. However, equally important is the quality control performed during the construction phase.

Interaction vesicles as emerging mediators of host‐pathogen molecular crosstalk and their implications for infection dynamics

open access: yesFEBS Letters, EarlyView.
Interaction extracellular vesicles (iEVs) are hybrid vesicles formed through host‐pathogen communication. They facilitate immune evasion, transfer pathogens' molecules, increase host cell uptake, and enhance virulence. This Perspective article illustrates the multifunctional roles of iEVs and highlights their emerging relevance in infection dynamics ...

Simulating Realistic MRI variations to Improve Deep Learning model and visual explanations using GradCAM [PDF]

open access: yesarXiv, 2021
In the medical field, landmark detection in MRI plays an important role in reducing medical technician efforts in tasks like scan planning, image registration, etc. First, 88 landmarks spread across the brain anatomy in the three respective views -- sagittal, coronal, and axial are manually annotated, later guidelines from the expert clinical ...

STAT3 expression is reduced in cardiac pericytes in HFpEF and its loss reduces cellular adhesion and induces pericyte senescence

open access: yesFEBS Letters, EarlyView.
Heart failure with preserved ejection fraction (HFpEF) accounts for half of the heart failure cases. It is characterised by microvascular dysfunction, associated with reduced pericyte coverage and diminished STAT3 expression in pericytes. Loss of STAT3 impairs pericyte adhesion, promotes senescence, and activates a pro‐fibrotic gene program.

Ion channel function of polycystin‐2/polycystin‐1 heteromer revealed by structure‐guided mutagenesis

open access: yesFEBS Letters, EarlyView.
Mutations in polycystin‐1 (PC1) or polycystin‐2 (PC2) cause autosomal‐dominant polycystic kidney disease (ADPKD). We generated a novel gain‐of‐function PC2/PC1 heteromeric ion channel by mutating pore‐blocking residues. Moreover, we demonstrated that PC2 will preferentially assemble with PC1 to form heteromeric complexes when PC1 is co‐expressed ...

AI Technicians: Developing Rapid Occupational Training Methods for a Competitive AI Workforce [PDF]

open access: yes
The accelerating pace of developments in Artificial Intelligence~(AI) and the increasing role that technology plays in society necessitates substantial changes in the structure of the workforce. Besides scientists and engineers, there is a need for a very large workforce of competent AI technicians (i.e., maintainers, integrators) and users~(i.e ...
