Endogenous Amplification of Apoptosis via p53 Regulation using a Cascade Nanocatalytic Medicine
An engineered MOF nanomedicine (MAL) triggers cascade nanocatalysis in tumors, converting H2O2 and L‐Arg into cytotoxic peroxynitrite. This induces DNA damage, activates the p53 pathway, and amplifies mitochondrial apoptosis via the BAX/Bcl‐2/Caspase‐3 axis, effectively inhibiting colorectal cancer growth with high biocompatibility.

To address multidrug‐resistant urinary tract infections (MDR‐UTIs), we developed Cu‐ZIF8‐Ru nanozyme featuring an asymmetric Cu─N─Ru catalytic site. A cascade‐responsive MXene@Cu‐MOF/GelMA hydrogel is engineered as a “skeleton–backpack” platform for extensive tracheal repair. The MXene framework scavenges postoperative ROS and converts NIR light into mild hyperthermia, while the Cu‐MOF component provides pH/NIR‐responsive Cu2+ dosing for infection control, angiogenesis, and chondrogenesis.
