A barley protoplast system has been established that supports replication of Rice hoja blanca tenuivirus (RHBV). Following polyethylene glycol-mediated RHBV inoculation of barley protoplasts, newly synthesized viral RNAs and proteins could be detected.

The rice stripe virus NS3 protein interferes with the MEL–SHMT1 module‐mediated defence response by competitively binding to the microtubule‐associated E3 ligase (MEL) substrate recognition site, thereby inhibiting MEL interacting with and ubiquitinating serine hydroxymethyltransferase (SHMT1).
