Bioinspired, Guanidinium, and Indole Modified Poly(glycidyl ether)s as Highly Efficient Vectors for Polyplex-Mediated Gene Delivery. [PDF]

open access: yesMacromol Rapid Commun
Guanidinium‐ and indole‐functionalized poly(glycidyl ether)s were developed as non‐viral gene delivery vectors with tunable hydrophilic–hydrophobic balance. All polymers formed stable DNA polyplexes, while moderate indole content yielded the highest transfection efficiency at low N/P ratios.

Dual-Functional Polyphosphoesters for Gene Delivery: Synergistic Effects of Guanidinium and Hydrophobic Side Chains in Degradable Polymers. [PDF]

open access: yesAdv Healthc Mater
This study presents guanidinium‐ and indole‐functionalized polyphosphoesters as degradable, non‐viral gene delivery vectors. Through precise tuning of charge density and hydrophobicity, these polymers form stable polyplexes with low toxicity. Remarkably, minor structural changes yield up to 200‐fold differences in transfection efficiency, highlighting ...

Loading of Extracellular Vesicles with Nucleic Acids via Hybridization with Non-Lamellar Liquid Crystalline Lipid Nanoparticles. [PDF]

open access: yesAdv Sci (Weinh)
This study explores how lipid nanoparticles interact with extracellular vesicles, emphasizing the importance of lipid nanoparticle topology in facilitating a controlled hybridization process between the two nanocarriers. The resulting hybrid nanoparticles display extracellular vesicle membrane proteins on their surface, are loaded with the nucleic acid

Pulmonary siRNA Delivery with Sophisticated Amphiphilic Poly(Spermine Acrylamides) for the Treatment of Lung Fibrosis. [PDF]

open access: yesSmall
Amphiphilic, cationic poly(spermine acrylamides) are synthesized via facile radical polymerization of N‐acryloxysuccinimide and N‐decylacrylamide followed by post‐polymerization functionalization with spermine. In vitro 2D and 3D cell culture, ex vivo and in vivo experiments reveal superior properties of spermine‐copolymers in delivery of siRNA to lung

The Effect of Cryoprotectants and Storage Conditions on the Transfection Efficiency, Stability, and Safety of Lipid‐Based Nanoparticles for mRNA and DNA Delivery

open access: yesAdvanced Healthcare Materials, Volume 12, Issue 18, July 17, 2023., 2023
In this study, the impact of storage conditions and the use of cryoprotectants on the medium‐term preservation of lipid‐based nanocarriers is investigated. Nanoparticle stability appears to be directly related to temperature, use of cryoprotectants and type of encapsulated nucleic acid cargo. The effect of storage on the physicochemical characteristics,

PLA‐b‐SMA as an Amphiphilic Diblock Copolymer for Encapsulation of Lipophilic Cargo

open access: yesMacromolecular Chemistry and Physics, Volume 224, Issue 1, January 2023., 2023
The synthesis of poly(lactic acid)‐b‐poly(styrene‐alt‐maleic acid) (PLA‐b‐SMA) via sequential ring opening polymerization and reversible addition‐fragmentation chain transfer polymerization is presented. This polymer has a hydrophilic (SMA) and hydrophobic (PLA) segment, allowing it to undergo nanoprecipitation for the production of nanosphere and ...
