Adverse effects of tocolytic therapy
BJOG: An International Journal of Obstetrics & Gynaecology, 2005The rationale for using tocolytics in preterm labour is to enable transfer of the mother to a tertiary centre and to prolong pregnancy sufficiently so that glucocorticoids can be administered to the mother. There is little question that these short term objectives can be achieved with contemporary tocolytics.

Frequently asked questions on tocolytics
BJOG: An International Journal of Obstetrics & Gynaecology, 2005The majority of drugs that are used today for tocolysis have been developed for clinical conditions other than preterm labour. Obstetricians started to use these drugs as tocolytics because of their additional influence on uterine smooth muscle relaxation, accepting their cardiovascular (side) effects.

Oxytocics, Tocolytics, and Prostaglandins
Anesthesiology Clinics of North America, 1990Summary Tocolytic and oxytocic therapies are widely used in contemporary obstetric practice. The anesthesiologist is often called upon to participate in the care of patients who are receiving uterotonic medications. In addition to their effects on the myometrium, each of these drugs has direct and indirect effects on a variety of organ systems ...

To examine the effectiveness of any tocolytic compared with a placebo or no tocolytic for preterm labor.We checked MEDLINE (1966-1998) and the Cochrane Controlled Trials Register for articles, using the search terms "randomized controlled trial" (RCT), "preterm labor," "tocolysis," "betamimetics," "ritodrine," "terbutaline," "hexaprenaline ...

Risk–benefit effects of tocolytic therapy
Expert Opinion on Drug Safety, 2004Tocolytics are potent drugs that are used to interdict preterm labour. Although all of these agents have some side effects, if not frankly adverse effects under certain clinical situations, two of these drugs, the beta-mimetics and magnesium sulphate (MgSO(4)), have been found to have considerable potential for adverse maternal cardiovascular and ...

Tocolytic Therapy: A Meta-analysis and Decision Analysis
Obstetric Anesthesia Digest, 2009To determine the optimal first-line tocolytic agent for treatment of premature labor.We performed a quantitative analysis of randomized controlled trials of tocolysis, extracting data on maternal and neonatal outcomes, and pooling rates for each outcome across trials by treatment. Outcomes were delay of delivery for 48 hours, 7 days, and until 37 weeks;

Tocolytic therapy represents the final obstetric end point in the prevention of premature births. For the past two decades, betamimetic therapy has been the mainstay of tocolytic therapy. Newer agents, indomethacin and nifedipine, offer new hope in the treatment of premature labor.

Magnesium sulfate as a tocolytic agent
American Journal of Obstetrics and Gynecology, 1983Magnesium sulfate (MgSO4) has been successfully used to inhibit premature labor. A retrospective review was performed on the use of MgSO4 as a tocolytic agent at Memorial Hospital, Long Beach, California, during a 4-year period (1978-1982). Three hundred fifty-five patients with diagnoses of premature labor were treated with MgSO4 after transport from ...

Risk-Benefit Assessment of Tocolytic Drugs
Drug Safety, 1991beta 2-Mimetics are the principal agents used for myometrial relaxation. As all the available drugs also have beta 1-stimulant effects, the various side effects (cardiovascular, pulmonary and metabolic) require a critical consideration of the clinical indications, thorough supervision and combined therapeutic concepts.
