Inhibition of cyclin‐dependent kinases 12/13 using CT7439 as a treatment for colorectal cancer with CDK12 upregulation

open access: yesMolecular Oncology, EarlyView.
The proposed mechanism of action for the CDK12/13 inhibitor and cyclin K degrader, CT7439. CDK12/13 inhibition interrupts transcription elongation, leading to increased DNA damage that results in cell death. This agent is a potentially novel treatment option for patients with colorectal cancer. Created in BioRender. Cyclin‐dependent kinase (CDK) 12 and

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Astrocyte heterogeneity in brain metastases

open access: yesMolecular Oncology, EarlyView.
Astrocytes emerge as pivotal regulators of metastatic colonization, survival, immune remodeling, and therapy response associated with an increasing heterogeneity that requires spatially and longitudinally resolved approaches to uncover regulatory programs and guide context‐specific therapies.

From tumor‐centric to ecosystem‐based hypotheses in brain tumor research and care

open access: yesMolecular Oncology, EarlyView.
Primary brain tumors, whether in adults or children, present a major challenge because of their dramatic prognosis and the ongoing lack of efficient therapeutic approaches. In recent years, a shift has occurred from tumor‐centric concepts to a more holistic view of these tumors as dynamic ecosystems.

Preoperative circulating tumor cells integrated with imaging analysis for prognostic evaluation in head and neck squamous cell carcinoma

open access: yesMolecular Oncology, EarlyView.
Detecting circulating tumor cells (CTCs) in blood before surgery may help predict outcomes in patients with head and neck squamous cell carcinoma (HNSCC). Here, we show when combined with tumor size and lymph node involvement from routine imaging, CTC status identifies high‐risk patients with poorer survival—offering a simple, minimally invasive tool ...

Total Variation in Imaging [PDF]

open access: yes, 2011
The use of total variation as a regularization term in imaging problems was motivated by its ability to recover the image discontinuities. This is at the basis of his numerous applications to denoising, optical flow, stereo imaging and 3D surface reconstruction, segmentation, or interpolation, to mention some of them.

Total Generalized Variation

SIAM Journal on Imaging Sciences, 2010
The novel concept of total generalized variation of a function $u$ is introduced, and some of its essential properties are proved. Differently from the bounded variation seminorm, the new concept involves higher-order derivatives of $u$. Numerical examples illustrate the high quality of this functional as a regularization term for mathematical imaging ...

Properties of minimizers of the total variation and of the solutions of the total variation flow

2014 IEEE International Conference on Image Processing (ICIP), 2014
We review some results obtained in collaboration with Vicent Caselles (formerly with DTIC, U. Pompeu Fabra, Barcelona) and Matteo Novaga (U. Pisa) and recent extensions obtained by the first author on the properties of the jump sets of solutions of total variation minimizing problems, including some results on explicit solutions in the 1D and radial ...
