INtraVESical immunoTherapy (INVEST) prior to radical cystectomy for bladder cancer: trial protocol
Background High‐risk non‐muscle‐invasive bladder cancer (HRNMIBC) has a variable prognosis, managed predominantly by local surgical resection and intravesical Bacillus Calmette–Guérin (BCG), or radical cystectomy (RC). Current treatments are poorly tolerated, have supply limitations and often fail to control the disease.

ABSTRACT Drug hypersensitivity reactions (DHRs) are common in children and are diagnostically challenging, with cutaneous manifestations being the most common presentation. Patch testing (PT) is a well‐established tool for investigating delayed T‐cell‐mediated reactions and is widely used in patients with contact dermatitis.

Objective The American College of Rheumatology recommends HLA‐B*58:01 allele testing before the initiation of allopurinol, specifically among Asian and African American/Black patients, due to their increased risk for severe hypersensitivity reactions. However, testing rates remain low at many health care facilities.

ABSTRACT Background T‐cell–engaging bispecific antibodies are increasingly utilised in haematological malignancies and are beginning to be adopted in solid‐tumour treatment protocols. Cytokine release syndrome and neurotoxicity are well recognised, but target‐specific dermatological adverse event reporting patterns remain poorly defined.

A Challenging Case of Acute Methotrexate Toxicity
ABSTRACT Catastrophic methotrexate toxicity, characterized by life‐threatening pancytopenia and neutropenic enterocolitis, may occur even after a single low therapeutic dose, particularly in patients with renal impairment or dehydration. Early recognition and prompt multidisciplinary management are essential to improve outcomes.

ABSTRACT Stevens‐Johnson syndrome/toxic epidermal necrolysis (SJS/TEN) overlap is a rare, life‐threatening mucocutaneous reaction typically induced by medications. Reports from Central America remain scarce. We present two contrasting cases of SJS/TEN overlap from Nicaragua.

Wetzel et al. outline how individual omics methods contribute to the diagnosis of patients with rare, and particularly mitochondrial diseases, with a focus on how spatial proteomics is joining this multi‐omics stack. ABSTRACT Proteomics by mass spectrometry has rapidly matured from a niche method into a standard tool.
