Modeling Protein–Protein and Protein–Ligand Interactions by the ClusPro Team in CASP16
ABSTRACT In the CASP16 experiment, our team employed hybrid computational strategies to predict both protein–protein and protein–ligand complex structures. For protein–protein docking, we combined physics‐based sampling—using ClusPro FFT docking and molecular dynamics—with AlphaFold (AF)‐based sampling, followed by AF‐based refinement.

Harnessing synthetic key receptor modulators, this study presents a breakthrough in Wnt/β‐catenin regulation. The FZD agonist RRP‐pbFn bypasses LRP5/6 to drive organoid expansion and tissue regeneration, while the LRP antagonist RRP‐Dkk1c eradicates tumors. These tools enable precise pathway control, advancing regenerative and cancer therapies.

A bionic nanomedicine system with core–shell structure (Nm‐pAu/CP/KU) was constructed. PEGylated gold nanoparticles (pAu) were loaded with cisplatin (CP) and a radiochemotherapy sensitizer (KU‐60019) to form the core pAu/CP/KU, and homologous nasopharyngeal carcinoma cell membrane vesicles (Nm) were used as the shell.
