Effects of pyrazole on rat liver tryptophan oxygenase

Abstract The effects of pyrazole administration on rat liver tryptophan oxygenase have been studied both under basal conditions and after induction by cortisol or activation by tryptophan. Pyrazole administration is followed by a decrease of the basal holoenzyme and total enzyme activities.

Abstract: 11–Deoxycortisol (cortexolone) has previously been used as a glucocorticoid antagonist in vitro and in adrenalectomized rats. Antiglucocorticoid properties of 11–deoxycortisol in intact rats were examined by studying the effect of 11–deoxycortisol on the induction of hepatic tryptophan oxygenase (TO) by corticosterone.

Abstract A spectrophotofluorometric assay was used to measure tryptophan oxygenase activity in several species. The fluorescent assay depends on the conversion of the product of the reaction, N-formyl- l -kynurenine, to anthranilate by means of the coupling enzymes kynurenine formamidase and kynureninase.

Abstract Chemically oxidized, catalytically inactive, pseudomonad l -tryptophan-2,3-dioxygenase (EC 1.13.1.12) can be photoactivated aerobically as well as anaerobically by light of wavelength less than 360 nm. The substrate, l -tryptophan, must be present for photoactivation to proceed.

Ethanol-induced increase in liver tryptophan oxygenase activity in the starved rat: Evidence against tryptophan mediation

A single oral dose of ethanol (4.0 g/kg) increased the activity of liver tryptophan oxygenase in starved male rats. The peak increase of 340% for the total activity and 400% for the holoenzyme activity occurred 6 hr after ethanol administration.
