Tyrosinase Activated Melanoma Prodrugs

Anti-Cancer Agents in Medicinal Chemistry, 2009
Metastatic malignant melanoma remains a highly aggressive form of skin cancer for which no reliable methods for treatment exist. Given the increasing incidence of this cancer, considerable attention has focused on the development of new and improved methods for tackling this disease.

Tyrosinase inhibition and anti-melanin generation effect of cinnamamide analogues.

Bioorganic chemistry (Print), 2019
Abnormal melanogenesis results in excessive production of melanin, leading to pigmentation disorders. As a key and rate-limiting enzyme for melanogenesis, tyrosinase has been considered an important target for developing therapeutic agents of pigment ...

Tyrosinase scavenges tyrosyl radical

Biochemical and Biophysical Research Communications, 2003
Melanosomes scavenged tyrosyl radical that was generated by ultraviolet irradiation of tyrosine. Purified mushroom tyrosinase also removed tyrosyl radical in a dose-dependent manner. To elucidate the underlying mechanism, we analyzed the reaction of mushroom tyrosinase with tyrosyl radical generated by horseradish peroxidase and hydrogen peroxide ...

Synthesis, computational molecular docking analysis and effectiveness on tyrosinase inhibition of kojic acid derivatives.

Bioorganic chemistry (Print), 2019
Tyrosinase inhibitors have become increasingly important as whitening agents and for the treatment of pigmentary disorders. In this study, the synthesis of kojic acid derivatives having 2-substituted-3-hydroxy-6-hyroxymethyl/chloromethyl/methyl ...

Relationships of dietary flavonoid structure with its tyrosinase inhibitory activity and affinity

LWT, 2019
Flavonoids possess the inhibition ability against tyrosinase. Here, the effects of flavonoids structure on inhibitory activity and binding affinity of tyrosinase were investigated. The results showed that the hydroxy groups of 5C−OH, 4′C−OH and 5′C−OH of

Validation of medicinal herbs for anti-tyrosinase potential

Journal of Herbal Medicine, 2018
Tyrosinase is a key regulatory multifunctional enzyme containing copper that is responsible for the biosynthesis of melanin that determines the color of the skin. Accumulation of excessive melanin causes various dermatological disorders including melasma

Tissue adhesive, rapid forming, and sprayable ECM hydrogel via recombinant tyrosinase crosslinking.

Biomaterials, 2018
We report on a tissue adhesive hydrogel based on novel recombinant tyrosinase mediated crosslinking. The adhesive hydrogels were fabricated by the site-directed coupling of tyramine-conjugated hyaluronic acid (HA_t, 1% w/v) and gelatin (3% w/v) (HG_gel ...

Catalytic mechanism of tyrosinases

Tyrosinases (TYR) play a key role in melanin biosynthesis by catalyzing two reactions: monophenolase and diphenolase activities. Despite low amino acid sequence homology, TYRs from various organisms (from bacteria to humans) have similar active site architectures and catalytic mechanisms. The active site of the TYRs contains two copper ions coordinated

The inhibition of tyrosinase

Archives of Biochemistry and Biophysics, 1955
Abstract The inhibition of tyrosinase by halide salts and carboxylic acids has been studied at several pH values. The presence of the benzene nucleus complements the inhibitory characteristics of carboxylic acids, and the existence of a negatively charged group on the inhibitor is also shown to be a factor.
