An Extracellular Pore‑Targeting Peptide Defines a Designable Allosteric Site in TRPV2

Structure‐guided peptide engineering yields Depiv2, a highly potent and subtype‐selective TRPV2 inhibitor that binds to the extracellular pore and remodels it into a closed, non‐conductive state. Depiv2 suppresses pathological cardiac hypertrophy, establishing the TRPV2 outer pore as a designable interface for selective peptide modulation.

RBM25 Drives Hepatocellular Carcinoma Progression by Stabilizing YAP Through Regulating Oncogenic Splicing‐switch of MYPT1

This study establishes that the RBM25‐PRPF40A interaction modulates MYPT1 splicing, promoting the production of the oncogenic long isoform. This isoform stabilizes YAP by suppressing its phosphorylation and subsequent proteasomal degradation, ultimately accelerating tumor growth.

Lactylation‐Driven PROS1‐TYRO3‐CARF Signaling Promotes Therapy‐Induced Senescence Escape and Radioresistance in Meningioma

A glycolysis‐driven epigenetic switch promotes therapy resistance in meningioma. Increased lactate production enhances histone H3K27 lactylation, which activates PROS1 secretion. Secreted PROS1 engages TYRO3 signaling to degrade the senescence regulator CARF, allowing cancer cells to escape growth arrest.
