Mechanical Signatures of Nucleic Acid Trefoil Knot Topology

Single‐stranded DNA can be designed with sequences that self‐thread into topological knots. The exact same sequence can alternatively be turned into an unknotted pseudoknot. The topology of the resulting polymer is determined by applying force with optical tweezers, causing the pseudoknot to resolve but the knot to tighten.
