Discovery of Ro 41‐0960, a Non‐Steroidal Inhibitor of the Na+/K+‐ATPase

Ro 41‐0960 is a non‐steroidal Na+/K+‐ATPase inhibitor with low micromolar potency and a mixed inhibitory mechanism distinct from classical cardiotonic steroids, without acute pro‐arrhythmic effects in human cardiomyocytes. The Na+/K+‐ATPase (NKA) is a central regulator of cardiac ion homeostasis and a validated target for heart failure.

Common structural issues such as atomic overlaps, abnormal bond lengths, and ring‐bond intersections can be rapidly detected in the pairlist‐enabled and parallelized “checkstructure” command from CPPTRAJ, for example, the ~256 k atom 5a9z system can be checked in less than 2 s using 16 threads.

Evaluating the Solvent and pH Effect in the Inversion of the Circular Dichroism Signal of S‐Naproxen

This study provides a theoretical and experimental analysis of the electronic circular dichroism (ECD) spectrum inversion observed in S‐naproxen when deprotonated through pH control. ABSTRACT S‐Naproxen (NPX) exhibits an unusual inversion of its ECD spectrum upon solvent change, a phenomenon that this work analyzes both experimentally and theoretically.
