Restriction Fragment Length Polymorphism in Plants and Its Implications

Abstract

Botstein et al. (1980) were the first to outline convincingly how cloned pieces of eukaryotic DNA could be used as genetic markers. They pointed out that organisms differing in base sequence would be expected to yield fragments of different sizes when their DNA was digested with a suitable restriction enzyme. These DNA fragments could then be fractionated by size on an agarose gel and detected by hybridization with a cloned probe homologous to a portion of the fragment. Such differences in the size of restriction fragments were termed restriction fragment length polymorphisms (RFLPs), and they would be expected to arise from nucleotide changes in the DNA or by insertions, deletions, or other forms of rearrangement.
