The LINGO of remyelination

LINGO1 is a negative regulator of oligodendrocyte differentiation and myelination. As Lingo1-knockout mice do not show any apparent developmental or behavioural abnormalities, the authors set out to investigate the therapeutic potential of LINGO1 inhibition in demyelinating diseases using mice with myelin oligodendrocyte glycoprotein (MOG)-induced experimental autoimmune encephalomyelitis (EAE), an established animal model for MS.

First the authors showed that, after EAE induction, disease progression was slower in Lingo1-knockout mice than in wild-type animals. Electron micrographs of the lumbar spinal cord showed remyelination in EAE-induced, Lingo1-knockout mice, but demyelination in wild-type control mice.
