Applications of high-sensitivity flow cytometry for single mitochondria analysis [PDF]
线粒体是细胞生命活动的控制中心,它不仅是细胞的能量中心,而且在多种诱因引发的细胞凋亡信号转导中起着重要的调节作用。以线粒体为切入点的信号转导研究,无论对于揭示细胞凋亡发生过程的分子机制,还是研究小分子化合物诱导肿瘤细胞凋亡的作用机理,均具有重要价值。与全细胞分析相比,单个线粒体水平上的分析能够排除其他细胞器的干扰,揭示线粒体功能之间的异质性,并有助于线粒体亚类的发现。然而,由于线粒体尺寸微小,与其功能相关的生化物质含量低以及缺乏灵敏的检测手段等原因,单个线粒体水平上的检测一直以来都是一个富有挑战性的课题。

研究分析了欧洲第三代核电站(EPR,European Pressurized-water Reactor)核岛安全壳钢衬里的模块化吊装。首先研究了环形桁架在进行钢衬里吊装时最不利竖向荷载下的受力性能;其次对钢衬里吊装环形桁架进行了参数优化分析,研究环形桁架的不同参数变化对桁架性能的影响,确定出环形桁架的优化参数取值;最后对某核岛安全壳钢衬里穹顶吊装时的各项受力性能进行了研究,包括吊装受力分析、风荷载作用分析及吊耳节点有限元分析,并提出了一些参数的取值及吊装方案的合理建议,为工程施工设计提供了参考。
