目的:通过腹腔注射大剂量戊四氮建立急性大鼠癫痫模型,运用姜黄素进行干预,观察姜黄素对癫痫大鼠行为的影响和神经元的保护作用及其机制研究。 方法:实验选用健康雄性SD大鼠48只,完全随机分为:姜黄素组、对照组、模型组、实验组,每组12只大鼠,造模前进行相应的预干预,然后通过腹腔注射戊四氮建立急性癫痫动物模型。观察造模后各组大鼠行为学表现;尼氏染色、HE染色观察各组大脑病理变化;Tunnel染色观察各组大脑皮质神经元的凋亡情况;免疫组织化学方法观察姜黄素对海马组织或皮质部分LC3-Ⅱ、Beclin1表达的影响。

目的观察骨髓间充质干细胞移植对大鼠大脑中动脉栓塞后神经功能的影响及GDNF的表达。方法采用线栓法大鼠大脑中动脉栓塞(MCAO)模型,颈内动脉移植MSCs,Bederson神经功能评分观察大鼠神经功能,免疫组织化学和原位杂交技术检测GDNF表达。结果MSCs移植可显著提高大鼠局灶性脑缺血后神经功能,缺血周边区GDNF蛋白及mRNA表达水平均高于对照组。结论MSCs移植可促进神经功能恢复,可能通过增加GDNF的表达起到脑保护作用。
